پرایمینگ بذر موجب بهبود جوانهزنی بذر و مولفههای رشدی گیاهچهها تحت شرایط تنشهای زنده و غیرزنده میشود. مطالعه حاضر به منظور بررسی تأثیر بیوپرایمینگ بذر با تعدادی از جدایههای بومی قارچ Trichoderma harzianum روی شاخصهای جوانهزنی و بنیه بذر توده¬ی بذری زیره سبز انجام شد. همچنین تأثیر آنزیمهای خارج سلولی تولیدشده توسط این جدایهها بهعنوان الیسیتور بر تولید و انباشت متابولیتهای ثانویه در گیاهچههای حاصل از بذور بیوپرایم شده مورد بررسی قرار گرفت. نتایج نشان داد که تمامی جدایههای T. harzianum آنزیمهای خارج سلولی به عنوان اصلیترین آنزیمهای هیدرولیز کننده دیواره سلولی تولید میکنند. تجزیه و تحلیل فعالیت آنزیمهای مخرب دیواره سلولی نشان داد که حداکثر میزان فعالیت هر آنزیم برای جدایههای مختلف T. harzianum متفاوت بود. حداکثر سطح فعالیت آنزیمی در میان جدایههای T. harzianum برای آمیلاز از 7/197 تا 338 μg.ml-1، پروتئاز از 3/15 تا 7/26 μg.ml-1، زایلاناز از 685 تا 7/1238 μg.ml-1، سلولاز از 442 تا 3/723 μg.ml-1، پکتیناز از 1030 تا 7/1583 μg.ml-1، کیتیناز از 7/12 تا 7/20 μg.ml-1 و لیپاز از 3/40 تا 7/22 μg.ml-1 میباشد. بالاترین میزان تولید پروتئین خارج سلولی و فعالیت آنزیمی در جدایه TH7 مشاهده شد. نتایج آزمایش گلخانهای روی گیاهچه های حاصل از بذور بیوپرایم شده نشان داد که میزان شاخص بیماری نسبت به شاهد آلوده از حدود 7 تا 35 درصد کاهش یافته است. نتایج نشان داد که بیوپرایمینگ بذر زیره سبز بهطور قابلتوجهی روی شاخصهای بنیه و جوانهزنی بذر تأثیر گذاشته و موجب بهبود کیفیت و سلامت بذر و گیاهچه شد. همچنین، قارچ T. harzianum و آنزیمهای ترشحشده توسط آن به عنوان الیسیتور موجب افزایش تولید و انباشت متابولیتهای ثانویه در زیره سبز شدند. یافتههای این پژوهش نشان داد که بیوپرایمینگ بذر به ویژه با جدایههای T. harzianum ضمن بهبود کیفیت بذر و سلامت گیاهچه، موجب افزایش میزان تولید و انباشت متابولیتهای ثانویه میشود.

Seed priming improves seed germination and growth components of seedlings under biotic and abiotic stress conditions. This study was aimed at investigating the effects of seed biopriming with a number of native isolates of Trichoderma harzianum on the germination and vigor indices of seeds of cumin population. Also, the effects of extracellular enzymes produced by these isolates as elicitors on the production and accumulation of secondary metabolites in seedlings obtained from bioprimed seeds were studied. Analyzing the activity of CWDEs revealed that the highest activity of each enzyme was different for different isolates of T. harzianum. The maximum level of CWDEs activity were among T. harzianum isolates for amylase from 197.7 to 338 μg.ml-1, protease from 15.3 to 26.7 μg.ml-1, xylanase from 685 to 1238.7 μg.ml-1, cellulase from 442 to 723.3 μg.ml-1, pectinase from 1030 to 1583.7 μg.ml-1, chitinase from 12.7 to 20.7 μg.ml-1 and lipase from 40.3 to 22.7 μg.ml-1. The highest amount of extracellular protein production and enzymatic activity was observed in TH7 isolates. The results of greenhouse experiment in the seedlings obtained from bioprimed seeds showed that the level of disease index decreased from about 7 to 35% compared to the infected control. The results showed that the cumin seed biopriming significantly affected the germination and seed vigor indices and improved the quality and health of seeds and seedlings. Also, T. harzianum and the enzymes secreted by it as elicitors increased the expression of genes associated with the production and accumulation of secondary metabolites in cumin. The findings of this research showed that seed biopriming, especially with T. harzianum isolates while improving seed quality and seedling health to cause increase the production and accumulation of secondary metabolites.
