خلاصه یا چکیده

بیماری جاروک مكزيكن لايم، که توسط Candidatus Phytoplasma aurantifolia ایجاد میشود، مهمترین عامل محدودکننده تولید مكزيكن لايم در مناطق جنوبی ایران است. بنابراین پژوهش حاضر با هدف بررسی تغییرات برخی ویژگیهای بیوشیمیایی و غلظت عناصر غذایی در برگ نهالهای ریشهدار مکزیکنلایم آلودهشده با فیتوپلاسما و مکزیکنلایم سالم طی پیشرفت بیماری (330-90 روز) و امکان جداسازی نهالهای سالم و آلوده تحت شرایط گلخانهای پایهریزی شد. بدینمنظور، قلمههایی از نهال سالم مکزیکنلایم تهیه شد. پس از سه ماه، نیمی از قلمههای مکزیکنلایم با استفاده از پیوندک پوست از نهال آلوده به عامل بیماری فیتوپلاسما آلوده شدند. در ادامه تعدادی برگ از هر یک از شش نهال سالم و شش نهال آلوده در بازههای زمانی 90، 150، 210، 270 و 330 روز پس از آلودگی نمونهبرداری شد و مقدار رنگدانهها، پروتئین، پرولین، فنل کل، ظرفیت آنتیاکسیدانی، فعالیت آنزیم-های پراکسیداز و سوپراکسیددیسموتاز، کربوهیدرات محلول و نشاسته در این برگها اندازهگیری شد. علاوه بر این، غلظت عناصر غذایی شامل نیتروژن، فسفر، پتاسیم، کلسیم، منیزیم، بور، آهن، منگنز، روی و مس نیز طی پیشرفت بیماری در برگ و در پایان آزمایش در برگ، ساقه و ریشه گیاهان اندازه گیری شد. نتایج نشان داد که آلودگی با فیتوپلاسما منجر به کاهش چشمگیر رشد و نیز کاهش وزن وزن خشك بخش هوایی و ریشه نهالهای مکزیکنلایم شد.علاوه بر این، آلودگی غالباً منجر به کاهش مقدار رنگدانهها و افزایش معنیدار مقدار کربوهیدرات محلول و نشاسته در برگ و کاهش این دو ویژگی در ریشه شد. همچنین، آلودگی با فیتوپلاسما منجر به افزایش معنیدار فنل کل و ظرفیت آنتیاکسیدانی و فعالیت آنزیم پراکسیداز در برگ و ریشه گیاه میزبان شد. به هر حال فعالیت آنزیم سوپراکسید دیسموتاز در ریشه و برگ گیاهان آلوده در مقایسه با گیاهان سالم به ترتیب افزایش و کاهش یافت. همچنين، بررسی غلظت عناصر غذایی در برگ گیاهان نشان داد که آلودگی با فیتوپلاسما منجر به کاهش معنی دار نیتروژن (25%)، کلسیم (23%)، بور (38%) و منگنز (26%) در برگ گیاهان آلوده و منجر به افزایش معنی دار غلظت فسفر (17%)، پتاسیم (25%)، منیزیم (75%)، آهن (86%)، روی (31%) و مس (9%) در برگ گیاهان آلوده شد. همچنین در مقایسه با گیاهان سالم، غلظت همه ی عناصر مذکور به استثناء منگنز در ساقه گیاهان آلوده کاهش و غلظت همه عناصر فوق به استثناء کلسیم در ریشه گیاهان آلوده افزایش یافت. به علاوه، نتایج تجزیه به مولفههای اصلی نشان داد که بر اساس شاخصهای بیوشیمیایی اندازهگیری شده در برگ گیاهان سالم و آلوده طی پیشرفت بیماری، میتوان این گیاهان را از یکدیگر تفکیک نمود ولی تجزیه به مولفه های اصلی بر اساس غلظت عناصر غذایی قادر به جداسازی گیاهان آلوده و سالم در مراحل اولیه نمونه برداری (قبل از ظهور علايم ظاهری بیماری) نبود. بهطور کلی نتایج مطالعه حاضر بینشی جدید در مورد پاسخ مکزیکنلایم به عامل بیماری فیتوپلاسما طی پیشرفت بیماری فراهم نموده و در اتخاذ راهکارهایی در جهت بازتوانی و افزایش عمر اقتصادی درختان آلوده به جاروک می تواند مفید باشد. واژههاي كليدي: بیماری جاروک، تجزیه به مولفههای اصلی، ترکیبات بیوشیمیایی، عناصر غذایی

Witches’ broom disease of lime (WBDL), caused by ‘Candidatus Phytoplasma aurantifolia’, is major limiting factor for Mexican lime (Citrus aurantifolia Swingle) production in the southern of Iran. In this investigation, leaf nutrient and biochemical composition profiling of the healthy and infected rooted Mexican lime was monitored during WBDL progression (90-330 days) as well as to distinguish infected from healthy Mexican limes under greenhouse condition. For this aim, stem cuttings is taken from healthy Mexican lime. After three months, half of Mexican lime cutting were inoculated by bark-grafting from WBDL-affected Mexican. Six healthy and infected plants were sampled 90, 150, 210, 270 and 330 days after inoculation with Phytoplasma and then measured pigments, protein, proline, total phenol, antioxidant capacity, the activity of peroxidase and superoxide dismutase enzymes, soluble carbohydrate and starch content in leaves . The leaf nutrient including nitrogen, phosphorus, potassium, calcium, magnesium, boron, iron, manganese, zinc and copper were also measured during WBDL progression and at the end of the experiment in leaves, stems and roots of the plants. The results showed Phytoplasmal infection caused growth inhibition and reduced stem and root dry mater. Moreover, infection mostly caused to a obvious decrease in pigments and significantly increase in soluble carbohydrates and starch and the reduction of these two characteristics in the root. In Mexican lime leaves and roots, antioxidant capacity and phenol content, peroxidase activity increased after inoculation with Phytoplasma. Howevere, the activity of superoxide dismutase enzyme in the root and leaf of infected plants increased and decreased respectively in comparison with healthy plants. Moreover, infection caused to a noticeable decrease in the concentration of nitrogen (25%), calcium (23%), boron (38%), manganese (26%), and significant increase in phosphorus (17%), magnesium (75%), potasium (25%), iron (86%), zinc (31%), copper (9%) in the infected leaves. Also, compared to healthy plants, there was a greater concentration of elements in leaves (exception manganese) and root (exception calcium) of infected plant. In addition, the principal components analysis (PCA) revealed a clear distinction between the leaf biochemical profiles of healthy and infected plants during the progression of WBDL. However, the principal components analysis based on the concentration of nutrients could not distinction infected and healthy plants in the early stages of sampling (before the appearance of symptoms). In conclusion, this study provides new insights into the lime response to phytoplasma infection during the progression of WBDL and It can be useful in adopting strategies to improve and increase the economic efficiency of trees infected with WBDL.
