Considering the need of the community for protein substances and the role of unrivaled proteins of aquatic origin in preserving and maintaining the health of the community on the one hand, and the process of development of rainbow trout farming in the country and the need for access to genera free of certain diseases and in order to Stability of production, on the other hand, is indispensable for the establishment of an SPF production center in the country. In this regard, one of the most important factors affecting the sustainable production and sustainable development of aquaculture in the country is the occurrence of various diseases and economic losses due to their occurrence in aquaculture and breeding centers. Today, the production of progeny free pathogens or SPF (Specific Pathogen Free) is recognized as an important tool for reducing the economic losses caused by diseases. This study attempts to address the importance of notifiable diseases and their impact on health status and potential damages caused by their occurrence and prevalence in different populations of aquatic animals, and with the emphasis of international health organizations such as (OIE) ) In this national plan, the efforts of all project implementers and colleagues were to use the international protocols and scientific resources to focus on the latest Organization Protocols (OIE) the notifiable diseases in all populations Study the macro plan for accurate health monitoring and based on the latest and most advanced diagnostic methods were employed to detect mentioned diseases. Sampling methods, sample size estimation, diagnostic methods, and diagnostic validation methods were all based on the most recent official OIE protocols. In this regard, the efforts of all the administrators were to highlight the main priority of viral diseases such as IPN, IHN and VHS in the designated headroom and the main site of the SPF in Tonekabon. Accordingly, the main structure of the health monitoring plan was approved and implemented. Therefore, in this health plan, three deadly and warning signs of Viral Hemorrhagic Septicemia (VHS), Infectious Hematopoietic Necrosis (IHN) and Infectious Pancreatic Necrosis (IPN), which are the leading cause of death of trout Selection of populations of healthy breeders for the production of Specific Pathogen Free (SPF) by cell culture, RT-PCR and Indirect fluorescent antibody test (IFAT) in Rainbow trout farms that had no history of the disease and approved by the Iran Veterinary Organization. In this study seven healthy populations were diagnosed and transferred to the pre-quarantine center. Of the studied farms, three IHNV contaminated farms and one farm contaminated with VHSV and IPNV that were identified and were removed from selected farms. In the follow-up, the diseases were monitored in the quarantine section of the SPF center before reproduction, during the reproduction season as well as in the fry population produced, and a contaminated population of VHS was removed before the proliferation season. Another goal of this national plan was to identify the various viral diseases in rainbow trout in the country, as well as to use the Real-time PCR method to quickly and accurately detect the common viral diseases in the fish populations that held in the SPF National Plan. The results of this study, using a trial of healthy and suspected viral diseases by using Real-Time PCR in the stages of implementation of this national plan, showed that this method can detect pathogenic viruses in rainbow trout. It is well suited for it to be used as a complementary method for further tissue culture testing as well as for rapid diagnostic method in screening and monitoring program in the country. After the molecular experiments, the results of the contamination of the fields were identified for each of the three viruses, Viral haemorrhagic septicemia (VHS), Infectious pancreatic necrosis (IPN), Infectious hematopoietic disease (IHN). Based on studies and epidemiological and eco-epidemiological studies on warning diseases in this national plan, it is concluded that increased bio-security in the predecessor farm has no a significant difference (p> 0.05) on the increase of immunity and serum antibody in any fish, and this index is fairly equal in fish in different centers. Of course, the lysozyme index as a nonspecific immune index in the SPF primary site pre-production farm has significantly increased, suggesting that upgrading the biological level could reduce stress and shift energy towards cultivating operations. Also, the results of postseasonal sampling showed that most of the measured indices and even the size of fish in the main site of the SPF center in Tonekabon were significantly higher than those of the selected farms. Based on the results of this study, it seems that the maintenance, nutrition management, rearing and sanitation management in the SPF center has been in a way that has led to a gradual increase in the number of fish kept in the center compared with the same herd in selected farms based on indices measured from a better health status.
